Flow cytometric analysis of IL-1 RAcP (IL-1R3) expression on peripheral blood leucocytes. Whole blood was treated with 1× BD Pharm Lyse™ lysing solution (Cat. No. 555899) to remove erythrocytes. The remaining leucocytes were washed and then stained with either BD Horizon™ BV421 Mouse IgG1, κ Isotype Control (Cat. No. 562438; Left Plot) or BD OptiBuild™ BV421 Mouse Anti-Human IL-1 RAcP (IL-1R3) (Cat. No. 748107; Right Plot) at 1.0 μg/test. The pseudocolor dot plots showing IL-1 RAcP (IL-1R3) [or Ig isotype control] staining versus side light-scatter were derived from gated events with the forward and side light-scatter characteristics of viable leucocytes. Flow cytometry and data analysis were performed using a BD LSRFortessa™ Cell Analyzer System and FlowJo™ software. The above is qualification data only and does not represent a specific OptiBuild™ lot.

Recommended Assay Procedures
For optimal and reproducible results, BD Horizon Brilliant Stain Buffer should be used anytime two or more BD Horizon Brilliant dyes (including BD OptiBuild Brilliant reagents) are used in the same experiment. Fluorescent dye interactions may cause staining artifacts which may affect data interpretation. The BD Horizon Brilliant Stain Buffer was designed to minimize these interactions. More information can be found in the Technical Data Sheet of the BD Horizon Brilliant Stain Buffer (Cat. No. 563794).

- This antibody was developed for use in flow cytometry.

The 89412 monoclonal antibody specifically recognizes the Interleukin-1 Receptor Accessory Protein (IL-1RAcP) which is also known as Interleukin-1 Receptor 3 (IL-1R3). IL-1RAcP (IL-1R3) is a type I transmembrane glycoprotein that is encoded by IL1RAP (interleukin 1 receptor accessory protein) which belongs to the immunoglobulin gene superfamily (IgSF). IL-1RAcP (IL-1R3) is comprised of an extracellular domain with three IgC-like domains, followed by a transmembrane region, and a cytoplasmic region with a Toll/IL-1R (TIR) domain. IL-1RAcP (IL-1R3) is widely expressed at low levels. This coreceptor associates with other ligand-binding chains, IL-1R1/CD121a (for ligands IL-1a, IL-1b, or IL-38), IL-1R4 (for IL-33), or IL-1R6 (for IL-36a, b, g, or IL-38) to form functional signaling receptor complexes. The cellular response to these ligands can be modulated through a non-signaling association of IL-1RAcP (IL-1R3) with the membrane IL1R2/CD121b decoy receptor.
The antibody was conjugated to BD Horizon™ BV421 which is part of the BD Horizon Brilliant™ Violet family of dyes. With an Ex Max of 407-nm and Em Max at 421-nm, BD Horizon BV421 can be excited by the violet laser and detected in the standard Pacific Blue™ filter set (eg, 450/50-nm filter). BD Horizon BV421 conjugates are very bright, often exhibiting a 10 fold improvement in brightness compared to Pacific Blue conjugates.
