Flow cytometric analysis of CD105 expressed on mouse bEnd.3 cells. Cells from the mouse bEnd.3 (Endothelioma, ATCC CRL-2299) cell line were preincubated with Purified Rat Anti-Mouse CD16/CD32 antibody (Mouse BD Fc Block™) (Cat. No. 553141/553142). The cells were then stained with either BD Horizon™ BV786 Rat IgG2a, κ Isotype Control (Cat. No. 563335; dashed line histogram) or BD Horizon BV786 Rat Anti-Mouse CD105 antibody (Cat. No. 564746, solid line histogram). The fluorescence histogram showing CD105 expression (or Ig Isotype control staining) was derived from gated events with the forward and side light-scatter characteristics of viable bEnd.3 cells. Flow cytometric analysis was performed using a BD™ LSR II Flow Cytometer System.

Recommended Assay Procedures
For optimal and reproducible results, BD Horizon Brilliant Stain Buffer should be used anytime two or more BD Horizon Brilliant dyes (including BD Optibuild Brilliant reagents) are used in the same experiment. Fluorescent dye interactions may cause staining artifacts which may affect data interpretation. The BD Horizon Brilliant Stain Buffer was designed to minimize these interactions. More information can be found in the Technical Data Sheet of the BD Horizon Brilliant Stain Buffer (Cat. No. 563794/566349) or the BD Horizon Brilliant Stain Buffer Plus (Cat. No. 566385).

The MJ7/18 monoclonal antibody specifically binds to mouse CD105 (also known as endoglin) which is a homodimer of 90-kDa subunits and is predominantly expressed on vascular endothelial cells. High levels of mouse endoglin mRNA have been reported to be detectable in the ovary, uterus, NCTC-2071 fibroblasts, and to a lesser extent, in heart, muscle and stromal cells in connective tissue of various organs. Endoglin has been reported to play an essential role in embryonic angiogenesis. Both mouse and human endoglin display strong amino-acid sequence homology to the transmembrane and cytoplasmic regions of the type III TGF-ß receptor.
The antibody was conjugated to BD Horizon BV786 which is part of the BD Horizon Brilliant™ Violet family of dyes. This dye is a tandem fluorochrome of BD Horizon BV421 with an Ex Max of 405-nm and an acceptor dye with an Em Max at 786-nm. BD Horizon BV786 can be excited by the violet laser and detected in a filter used to detect Cy™7-like dyes (eg, 780/60-nm filter).
