Flow cytometric analysis of IgG expression on human peripheral blood lymphocytes. Human peripheral blood mononuclear cells (PBMC) were washed and cultured in complete tissue culture medium overnight in order to minimize nonspecific immunofluorescent staining. The cells were harvested and stained with PE Mouse anti-Human CD19 (Cat. No. 555413) and either Alexa Fluor® 700 Mouse anti-Human IgG antibody (Cat. No. 561296; Right Panel) or with an Alexa Fluor® 700 Mouse IgG1, κ Isotype Control (Cat. No. 557882; Left Panel). Two-color flow cytometric dot plots showing the correlated expression of cell surface IgG (or Ig isotype control staining) versus CD19 were derived from gated events with the forward and side light-scatter characteristics of viable lymphocytes. Flow cytometry was performed using a BD™ LSR II Flow Cytometer System.

Product Notices
- This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1 × 10^6 cells in a 100-µl experimental sample (a test).
- An isotype control should be used at the same concentration as the antibody of interest.
- Alexa Fluor® 700 has an adsorption maximum of ~700nm and a peak fluorescence emission of ~720nm. Before staining cells with this reagent, please confirm that your flow cytometer is capable of exciting the fluorochrome and discriminating the resulting fluorescence.

IgG is an important component of the humoral immune response, helping to control infection. IgG is produced by plasma B-cells and may be found in extracellular fluids, such as blood, lymph, peritoneal, and cerebrospinal fluids. IgG monomers consist of two light and two heavy chains containing two antigen binding sites. There are four IgG subclasses found in human, mouse and rat species, which include IgG1, IgG2, IgG3 and IgG4. The G18-145 monoclonal antibody specifically binds to the heavy chain of human immunoglobulin G subclasses: IgG1, IgG2, IgG3 and IgG4. The G18-145 antibody has been reported not to react with the heavy chains of other human immunoglobulin isotypes.
